Prototyping web based applications using XHTML, CSS and Javascript directly instead of traditional approaches like Visio, Illustrator or dedicate software has been gaining ground lately. Boxes and Arrows has a good write up describing this process and how to utilise it.
Also, at the 2008 @Media Ajax conference in London, Richard Rutter (Clearleft) gave a presentation about wireframing ajax interactions using a similar methodology.
